There are both postsynaptic and presynaptic receptors for both 5-ht1a, and 5-ht1b.
Activation of the post synaptic 5-ht1a/5-ht1b receptors is responsable for the antidepressant/antipanic effects.
If you activate the presynaptic (autoreceptors) then you shut off serotonin release. Ie you want to desensitize the presynaptic autoreceptors so that they don't get activated as much which results in more serotonin release to reach the post synaptic receptors.
People with depression have tons of really sensitive autoreceptors but their postsynaptic systems aren't working as well.
