No response to SAM-e 400mg/day- how do u titrate?

I been taking SAM-e 400 mg (2 nature made tablets) in the morning with a Centrum Multivitamin, Vitamin B6, & a folic acid 400mcg (100%RDA) for about 2 weeks now with little or no response.
I haven't gotten around to buying a sublingual vitamin b-12 pill yet, but am going to in the next few days.
So far I haven't had any response- I think I felt a bit better the 1st two days but then nothing after that.
How do you titrate the dose of SAM-e? (the Nature Made insert doesn't help that much). If you titrate to 800 mg are you supposed to take 2 pills in the early morning and 2 pills an hour before lunch or just take 4 pills all at once in the early morning (all on an empty stomach)?
Does anyone know the duration or 1/2 life of SAMe?
