I remember reading somewhere that inositol increases the sensitivity of the d2 receptors. Could this make a d2 agonist more effective.
Has anyone else read this / know where the study is?

Short term yes but...
the problem with any receptor exposed to agonists is the problem with down-regulation of receptors (desensitisation).
This process may vary between individuals. That is probably why some become addicts and others don't. This is also why so many drugs poop out.
