I just have started Anafranil few days ago and it's having a positive impact on my OCD. But it makes my pupils HUGE. I mean I look like I'm on meth, or something out of a horror movie. This is pretty embarrasing, especially at work.
Does this effect go away? I've had the same effect with SNRIs, but never took them long enough to see whether it goes away or not.

Clomipramine is a blocker of the following transporters:[4]
* Serotonin transporter (SERT) (Ki = 0.14 nM)
* Norepinephrine transporter (NET) (Ki = 54 nM)
* Dopamine transporter (DAT) (Ki = 3,020 nM)(wiki)
so the answer - your dopamine must be through the roof. This is normal and should subside somewhat with time (from my experience initial reaction is most drastic, then body tends to get used to it). But I would be less worried about it, just tell your colleagues, "don't mind my eyes (pupils), it's the medication."
Really few people pay that much attention to this. I think it it is mostly how one handles the self.

Higher numbers indicate lower affinity for the transporter. Lower numbers means that the drug binds to the site at a lower drug concentration.
Clomipramine has most of its affect on the serotonin transporter. It has negligable effect on the dopamine transporter.

you're correct! I've overlooked something.
http://books.google.com/books?id=9WfcTkROX_0C&pg=PA22States that the higher Ki value, the lower affinity (binding capacity).

The pupil dilation, medical term mydriasis, is due to the anticholinergic side-effects in the first place, but also a possible serotonergic side-effect. I think it could get better over time. I'm on Anafranil myself and sunlight can be very harsh for my eyes...
